O.K., if money is no object, you can easily get any of the masteries. I wanted to go elixir, but the general consensus was that it would take a couple of BM runs and my guild was not at the level of getting the Master's Key yet (so we weren't running BM).
So, I ran up to Netherstorm and handed over four Primal Mights and got the Transmute Mastery, figuring I could just then pay 150 gold to "unlearn" it and then get the Elixir Mastery. My understanding is that you do not have to do the quest for the second mastery if you get the first and drop it. Primal Mights go for between 60 and 90g on my server, so as you can see it costs a bit to do it this way, but for me its only a few hours of dailies, flower picking, and fishing.
Its more the opportunity cost because I can fish up the motes of water for the Primal Water needed to transmute a Primal Might, and of course even then the primals needed to transmute a Might on my server net at least 30g profit after the transmute if I sell the Might for 90g.
By the way, I wound up sticking with Transmute and am not sorry, but I can always switch if I decide to.
