Just started to play again and not sure how this whole glyph business work. What are they and how do I go about getting them? I am a lvl 42 Ret Pally. WHat would be the best Glyph for me to use?
All glyphs are made by Scribes, people who have taken the Inscription profession. The materials needed to make them also come from the Inscription profession, so really -- but best place to get them is straight off the AH.
As for what glyph.. none are absolutly game-breaking -- evaluate how you play and go from there.

http://wow.allakhazam.com/wiki/glyph_(wow) an incomplete list i think but gives you a rough idea what should be available to use , as to which is best it depends on what you think will be of use or that others say are worth using as i am not a Pally.
I would suggest if you are in a guild or can find someone willing to make them if you supply mats it may be a cheaper and better option than heading to the Auction House at the moment.

Pally Glyphs
Glyph of Blessing of Might no longer seems to work (for me) anymore. When I cast it on myself, it still only lasts 10 minutes. It used to work. It would give me 30 minutes.
If you want to equip them, you need to be near a Lexicon of Power (I think that's what it's called). You may need to ask the guard where it is. Some places, the guard will have an option for the device itself while others you have to ask for the inscription trainer.

I just read that the glyph works with BoM 1-5 but once you get BoM 6, at level 54, it stops working.

It's specifically rank 6. All other ranks work. I think the next rank is at 60, so.. stock up on a few symbols and cast greater for a while.
